Nosocomial infections caused by multidrug-resistant Pseudomonas aeruginosa remain a major clinical concern and require exploration of alternative antibacterial sources. This study evaluated the antibacterial potential of candidate Bacillus sp. isolated from the rhizosphere of De Djawatan Forest, Banyuwangi, Indonesia. Soil samples were collected from five locations, and 13 candidate Bacillus sp. isolates were obtained. Antagonistic activity against P. aeruginosa was assessed using the dual culture method, while the filtrate of the most active isolate was evaluated using a disk diffusion assay at different concentrations. GC-MS analysis was performed to characterize the chemical profile of the active filtrate. Candidate Bacillus sp. 8 showed the highest inhibition zone among the isolates, with a mean diameter of 19.75 mm. Its ethyl acetate filtrate retained inhibitory activity at concentrations as low as 5% (v/v). GC-MS profiling detected 13 compounds, with the highest relative abundance observed for hexadecanoic acid, methyl ester (65.8%) and octadecanoic acid, methyl ester (7.7%). These findings indicate that candidate Bacillus sp. from the De Djawatan rhizosphere has in vitro antibacterial activity against P. aeruginosa, although further molecular identification, compound isolation, and bioactivity validation are required.
